What PE-22-28 is
PE-22-28 is a short peptide analog of spadin, studied for rapid antidepressant-like effects in animal models. It drew interest because blocking its target produced fast-onset effects in rodents, unlike conventional antidepressants that take weeks. It has never been tested in humans.

How PE-22-28 works
It blocks TREK-1, a two-pore-domain potassium channel expressed in the brain that is implicated in mood regulation. Mice lacking TREK-1 show an antidepressant-resistant phenotype, and blocking the channel in animal models produced rapid antidepressant-like behavior. Whether this translates to humans is unproven.

A worked example
A 10 mg vial reconstituted with 2 mL of bacteriostatic water gives 5 mg/mL. A 500 mcg dose is then 0.1 mL — about 10 units on a 100-unit insulin syringe, giving roughly 20 doses per vial before accounting for residual volume left in the vial and syringe.
